Antioquía fue fundada en el año 300 a. C. por Seleuco I. Fue la capital del reino seléucida antes de convertirse en la de la provincia romana de Siria. Siria fue anexada por Pompeyo en el 64 aC Bajo el Imperio, Antioquía era una de las ciudades más pobladas del Imperio con 200.000 a 300.000 habitantes. Importante metrópolis del Oriente romano, también fue una ciudad amenazada por invasiones. Los partos y luego los sasánidas amenazaron o se apoderaron de la ciudad. Fue cristianizada muy tempranamente como Alejandría. La ciudad, cosmopolita, era el punto final de todas las caravanas de Oriente.
